through the day, throughout the day が一日中です。それに対して during the day は、その日の間ということなので、同じことが一日中ずっと続いているとか、同じことを一日中ずっとしているという訳ではありません。その日のうちにあること、することであれば during を使います。
I climbed Mt. Fuji yesterday. I kept moving through the day. I was hungry throughout the day. I ate ten rice balls during the day. I took many pictures and sent them to my friends during the day.
